Anna Ishii was born in Tokyo, Japan. She has an elder brother, a younger brother, and a younger sister named Chino Ishii, who is currently listed under Kizzy, an all-female dance group under LDH management, composed of EXPG students.

She started dancing when she was in her second year of primary school at her mother's encouragement. Ishii was later scouted to join the Exile Professional Gym in her fifth grade when she took part in a dance competition. She debuted as a model in the U-15 fashion magazine nico puchi in 2010 before joining E-girls.

In April 2012, Anna Ishii debuted as an actress with a cameo appearance in the drama Shiritsu Bakaleya Koukou. She continued to strengthen her acting skills in diverse roles such as Koibumi Biyori (2014), Solomon's Prejury (2015), and Girls Step (2015) which earned her the 58th Blue Ribbon Best New Comer Award.

During 2016, Ishii won the 5th Confidence Drama New Comer Award for her portrayal as Nagisa Arima in the drama Aogeba Tōtoshi. Not soon after, she starred in the live-action Your Lie in April as Tsubaki Sawabe.

A year later, Anna Ishii took on the role of Ayama Gotō in the drama Okāsan, Musume wo Yamete ii desu ka?, as well as Uso nante Hitotsu mo nai no, Parallel World, and the live-action version of The Anthem of the Heart (2017)

Currently, her most recent films and dramas include, We are Rockets! (2018), Utamonogatari "Kuu" (2018), and Love Stoppage Time (2019)
